Research Interests:
My overall research interests are in the biochemistry and molecular biology of enzyme-catalysed reactions and protein purification. During this period we established a methodology and I was initially pursued an investigation of the basic mechanisms of a novel trans-cis 2,4-DCA isomerase involved in the biosynthesis of the benzopyrone nucleus of coumarins. The benzopyrone nucleus is formed from the spontaneous ring closure of the cis forms of 2-hydroxycinnamic acids derived from L-phenylalanine. Following determination of L-phenylalanine to trans cinnamic acid, 2-hydroxylation and trans-cis isomerisation of the double bond of the propenoic acid side chain must occur prior to ring closure. Many plants contain coumarin natural products, some of which possess biological activity, and some of which are used in phototherapy of skin diseases. Medically, coumarin glycosides have been shown to have blood-thinning, antimicrobial agents, anti-fungicidal, and anti-tumor activities. Dicumarol, a coumarin glycoside known as warfarin, is commonly used oral anticoagulant medication.
