Implementing a platform for high-throughput proteomics at UMB’s new Mass Spectroscopy instruments

One important corner stone in biotechnology is the availability of relevant, efficient and accurate measurements, and the ability to relate such measurements to other information in a systematic way.
Mass spectroscopy such as Maldi-TOF-MS (Matrix assisted laser desorption/ionization time-of-flight mass spectroscopy) and LC-MS (Liquid chromatography mass spectroscopy) can give very useful information about the composition of biological samples.
At UMB and campus Ås, recent years have seen a substantial investment in mass spectroscopy instruments. It is important that these investments in proteomics be used as widely and efficiently as possible. In order to do that, one must ensure that measurements can be taken easily and with good repeatability, and that methodology for treating and analysing the measurements is readily available. Developing a platform for high-throughput mass spectroscopy will facilitate future research in a wide range of animal and food topics.
MS instruments are expensive, and their potential as rapid and versatile measurement systems are not fully utilised. Measurements from MS instruments are today used more as qualitative measurements than quantitative. This makes predictive and other data analysis difficult.
The main objective of the project is to develop a platform for more efficient utilisation of several advanced mass spectorscopic instruments recently acquired at UMB, combined with modern data analysis. This can be divided into four subgoals:
1. Develop procedures for robust optimisation of instruments.
2. Test, tune and develop preprocessing methodology for MS measurements.
3. Investigate the effect of interactions in samples, and develop methodology for detecting and possibly correcting for interactions.
4. Develop the use of 2-way and N-way methods for data analysis.
